'U' uses gene therapy to treat high cholesterol

Researchers at the University's
Howard Hughes Medical Institute
(HHMI) have successfully begun to
use gene therapy to treat familial
hypercholesterolemia (FH), a lethal
form of high cholesterol.
Patients with FH lack a gene which
is responsible for the production of a
natural cholesterol-lowering protein.

The lethal form of FH affects one in a
million Americans.
A less severe form which leads to
premature heart attacks affects nearly
one in 500 Americans.
The procedure began June 5 with
surgery to remove a small portion of
liver tissue from a patient with FH.
This liver tissue was then geneti-
cally treated and the transformed cells
were re-infused into the patient's liver

three days later.
During those three days, the liver
tissue was broken down into individual
cells and exposed to a harmless virus
that caries the cholesterol-lowering
gene.
The virus was permitted to infect
the cells and use the cell's protein-
making system to manufacture the cho-
lesterol-lowering protein.
Now that the cells have been im-

planted into the liver again, it is hoped
that they will grow and continue to
produce the protein.
"We are pleased with how things
are going so far and are particularly
optimistic about the impact of this
therapy on the patient," said James
Wilson, the project's principal investi-
gator.
The use of gene therapy to treat
lethal diseases is a relatively new tech-

nique.
"Gene therapy has the potential to
significantly alter the way we approach
a number of inherited conditions and
other diseases which involve genetic
changes," said George Zuidema, vice
provost for medical affairs.
He added, "We are excited that the
Medical Center has taken another step
in this new frontier of medicine."
