Protein Regions Aid Insulin Action particular protein region seems to play a key role in the cell's response to in- sulin, according to a Weiz- mann Institute study described in the July 28, 1995, issue of the Journal of Biological Chemistry. Because loss of response to in- sulin is one of the causes of dia- betes, this finding may some day contribute to the development of new ways for treating this dis- ease. Insulin is a hormone formed in the pancreas and released into the bloodstream in order to pro- mote the uptake of glucose from the blood by the body cells. Dia- betes, which affects millions of people worldwide and is the third leading cause of death, is caused either by insufficient insulin pro- duction or by a lack of proper re- sponse to the insulin on the part of target tissues. The new study has pinpointed a region in the target cells that is necessary for a normal response to insulin. Cells sense the presence of in- sulin via specialized receptors that protrude through cell mem- branes and initiate biochemical changes inside the cell. The Weizmann team, headed by Pro- fessor Yehiel Zick of the depart- ment of chemical immunology, deleted a region of the cell pro- tein IRS-1, which is a biochemi- cal link in the insulin signaling pathways, and found that insulin signaling was disrupted. This re- gion, called the plackstrin ho- mology domain, plays an important role in insulin action. This conclusion is supported by similar findings in a Harvard Medical School-U.S. National In- stitutes of Health study report- ed in the May 19 issue of the Journal of Biological Chemistry. Professor Zick's team, which included departmental colleague Dr. Daniel Schindler and Ph.D. students Dr. Hedva Voliovitch and Yaron Hadari, collaborated with Drs. Simeon Taylor and Domenico Accili. The plackstrin homology do- main contains.about 120 amino acids grouped into six main blocks.
